The 31-year-old South Africa batsman hit 135 not out to help his side bounce back from 39 for three as they attempted to overhaul a first-innings deficit of 178.
Championship debutant Luke Fletcher took two out of the three wickets to fall in the day, trapping both Tom Smith and Francois du Plessis lbw.
Paul Horton was caught behind by stand-in wicketkeeper Bilal Shafayat off the bowling of Ryan Sidebottom.
But Prince shared an unbroken fourth-wicket partnership of 191 with Mark Chilton (56 not out) to help his side to 230 for three declared. Both sides agreed to shake hands just before 5pm.
Michael Carberry and James Adams scored half-centuries as Hampshire's LV County Championship match with Sussex petered out into a dull draw at the Rose Bowl.
Sussex, 120 behind overnight in their first innings, chose to bat on until they were all out in a bid for batting bonus points.
Hampshire took a 14-run lead into their second innings but had no option but to bat through the last two sessions to render the last day of a rain-hit match meaningless.
Sussex resumed on 230 for six in reply to Hampshire's first-innings total of 350 for eight declared and went on to make 336, with wicketkeeper Matt Prior scoring 140.
Damien Wright (42) and Ragheb Aga (24) put on 55 for the ninth wicket. Carberry then scored 72 and Adams an unbeaten 55 as Hampshire closed on 127 for two.
