Wicketkeeper Prior augmented the hundreds made by Paul Collingwood and Alastair Cook as England racked up 467 for five by midway through the evening session.
Prior (73no) made 72 out of a hundred stand which came up in only 113 balls, having hit nine fours in his 55-ball 50.
He had a perfect opportunity to demonstrate his attacking style - and made no mistake with a succession of crisp cover-drives and clubbed pulls.
A more workmanlike Collingwood (111) had earlier made West Indies pay for missed chances as he posted his fourth Test hundred.
He was twice dropped badly in the 30s and went on to share a stand of 144 with Bell (84no), helping to keep the pressure on for batting positions in an England middle order due to be reinforced by the returns from injury of Michael Vaughan and Andrew Flintoff at Headingley next week.
