Playing one of their two games in hand over leaders Wolves and second-placed Birmingham, the Royals laboured for much of the match at the Keepmoat Stadium.
However striker Dave Kitson, back at the club after an unsuccessful move to Stoke last summer, provided a crucial winner eight minutes from time, curling the ball home from Chris Armstrong's low centre.
Play-off hopefuls Ipswich and Burnley drew 1-1 at Portman Road.
While the Clarets are currently fifth, Ipswich have only an outside chance of reaching the play-offs and their hopes looked bleak when Wade Elliott scored for Burnley in the first half. But Ipswich took a point when on-loan forward Giovani Dos Santos scored in the 73rd minute.
Swansea let slip the chance to steal a march on their fellow play-off chasers when they lost 1-0 at QPR.
The Welsh side had their chances but Mikele Leigertwood's goal on the half-hour mark proved their undoing at Loftus Road.
Barnsley boosted their survival hopes and climbed out of the bottom three with a commendable 3-1 win at home to Crystal Palace.
Shefki Kuqi had given Neil Warnock's side an eighth-minute lead but Jamal Campbell-Ryce levelled in the 23rd minute and Lee Hills' own goal early in the second half put the Tykes ahead.
And Michael Mifsud wrapped the win up in the 73rd minute when he steered the ball past Julian Speroni.
