The Spaniard stayed out of trouble on the 143 kilometre run from Etampes to the French capital which culminated in several circuits around the Champs-Elysees.
It was enough to give Sastre overall victory by 58 seconds ahead of Australian Cadel Evans, making him the third successive Spaniard to win the Tour after Oscar Pereiro in 2006 and Alberto Contador in 2007.
Bernard Kohl, the King of the Mountains, took the third step on the podium.
Gert Steegmans won the stage for Quick Step after holding off a late charge from Gerald Ciolek to prevent Team Columbia from taking a sixth stage victory in the Tour.
Oscar Freire finished third behind Steegmans and Ciolek to safely secure the green jersey for the winner of the points competition.
Any chance of a successful escape ended when Alexandre Botcharov was swallowed up by Team Garmin Chipotle with 6km left.
As convention dictates, the yellow jersey was not attacked in the dash up and down the Champs-Elysees.
