After skipper Anil Kumble won the toss and elected to bat first in Adelaide, India progressed to 309 for five - with Tendulkar unbeaten on 124.
Opener Virender Sehwag also contributed 63, while Brett Lee and Mitchell Johnson picked up two wickets each for the hosts.
Having named five bowlers in the side - they brought in spinner Harbhajan Singh for Wasim Jaffer - India needed their star batsmen to score the bulk of the runs.
They were helped by some weak fielding by the Australians, but Sehwag, VVS Laxman (51) and Tendulkar showed character as well as class.
