Adam Johnson and Fraizer Campbell struck stunning first-half goals at Estadio La Rosaleda to shock the South Americans, who had senior players with World Cup experience in their ranks.
Pearce's youngsters were then pegged back when Ecuador struck twice in quick succession just before the hour mark, leaving England hanging on to extend their unbeaten run to 16 matches. That record went when Manchester City striker Felipe Caicedo struck the winner with eight minutes remaining.
Apart from a penalty shoot-out loss to Holland in 2007, the under-21s had not tasted outright defeat in 28 games - but Pearce wanted a genuine test for his squad.
Johnson opened the scoring in the 14th minute with a stunning individual strike. The Middlesbrough winger took advantage of a Segundo Castillo mistake by flicking the ball over the Everton midfielder, then surging into the box and doing the same to Deison Mendez.
He still required a finish and lashed a right-footed volley past Cevallos.
Campbell's goal came in the 42nd minute, a strike to finish off a rapid counter-attack. Micah Richards had cleared a corner then Tom Huddlestone lofted a ball over Ecuador's defence. Michael Kightly shifted the ball from left to right and Campbell took his finish first time, leaving Cevallos with no chance.
Ecuador were back on level terms with two goals in as many minutes after the interval. The first came in the 53rd minute when Edison clipped the ball over Joe Mattock, with Guerron cutting inside and slamming his finish through Tom Heaton's legs.
Pablo Palcios then raced through, behind England's defence, and finished calmly.
Caicedo's moment came with eight minutes remaining. He received the ball in the area, shifted it to his left foot and smashed it into Heaton's near post.
