The Warriors recovered from 10-0 down to run out 28-10 winners in front of a bumper 20,295 crowd at the newly-renamed DW Stadium.
Rising star Sam Tomkins capped the comeback and a superb individual performance by finishing a quick counter-attack for Wigan's fourth try late on.
George Carmont had begun the fightback before half-time in response to Danny McGuire and Scott Donald tries.
Harrison Hansen and Joel Tomkins also touched down for the hosts as Brian Noble's men won for a fifth game in succession.
Noble said: "I thought the second half was up there with what we have done because of the crowd, the emotion and the fact we were playing the champion team.
"I thought they punched holes in us in the first half, they were all over us.
"But, boy, did we respond. We had energy, we had vigour, we were meeting their bigger blokes as we should have done in the first half.
"We also looked after the ball a bit better, forced a few errors and we know we will score points if we get to the right end of the field."
The result ended Leeds' run of six consecutive victories and denied them the chance to reclaim top spot in the table from St Helens.
