The 29-year-old former Wakefield and Castleford winger, who made the move to the capital at the end of the 2003 campaign, has played in every season since Super League kicked off in 1996.
"I'm happy to commit myself to Harlequins Rugby League and to finish my career here," said Wells, who is currently sidelined with a knee injury.
"They have been very good to me over the past five years. We're a family club with real ambitions, and I'm glad to be a part of it.
"I think we all see the present as a work in progress, and I'm really excited about being here when we really start to see the benefit of everyone's hard work.
"Now I'm just looking forward to getting this brace off my leg and back onto the field to contribute again - players make very bad spectators."
The news comes just four days after Harlequins coach Brian McDermott pledged his future to the club by signing a new three-year deal.
