The first set was level at 2-2 after the opening four games, but Serena picked up the next four to wrap up the opening set on the Arthur Ashe court in 27 minutes after converting two of three break opportunities.
Two-time champion Serena continued to dominate the contest and wrapped up the win in 60 minutes to set up a repeat of July's Wimbledon final.
Earlier, two-time US Open champion Venus also dropped just four games in beating Poland's Agnieszka Radwanska in straight sets.
Also on Monday, sixth-seeded Dinara Safina of Russia reached the quarter-finals for the second time in three years with a 7-5 6-0 victory over Anna-Lena Groenefeld.
A winner in Berlin, Los Angeles and Canada and a silver medallist at the Beijing Olympics, the 22-year-old will next face 16th seed Flavia Pennetta of Italy.
Pennetta advanced to her first quarter-final at a grand slam by downing a struggling Amelie Mauresmo 6-3 6-0.
Mauresmo committed 14 double faults and was broken seven times and failed to make it past the fourth round of a grand slam for a seventh consecutive time.
